There's not really anything worth pondering here. No matter what they do, it will assuredly be the superior version. Doing the bare minimum of all DLC and/or minor graphical enhancements will already make it "superior."
In approximately one year, Sakurai and a tiny DLC team created seven additional characters and eight stages.
If Sakurai was so inclined, he and his team could easily release Smash NX as a launch title next March with a handful of exclusive characters and stages. Plus, its a retail product, so it would have a much larger team than the DLC did.
If there is an NX version, all they would have to do is add Ice Climbers and Wolf and I would buy it again.

There will be a need for an actual Smash 5 in the near future, one that not only fixes the mistakes of Smash 4's gameplay but also improves on what it did right. As (Ultra) Street Fighter IV & Ultimate Marvel vs. Capcom 3 proved, you can't support the same game forever without the audience eventually growing bored of it. I wouldn't be surprised if they made amiibos for the alt costumes in Smash 4 like female Robin, male WFT, Wireframe Little Mac, Alph, Koopalings, etc

Note that our only info on this game is unofficial (Serkan Toto, Emily Rogers). We don't actually know that Nintendo will market it as a port of Smash Brothers for Wii U. It's actually rather unlikely (Smash Brothers for Wii U for NX...lulz).
Even if they are porting the Wii U game as a starting point, that doesn't mean that the product they release will be just the Wii U game on a different platform. In my opinion, this will be presented as a new Smash Brothers for the NX even if it proves to be more of a straight sequel than Melee/Brawl.
Also it's worth noting that the NX Smash is at least a year or so away. We got 6 DLC characters for Smash Wii U in about that time.
Porting will require effort but the assets aren't terribly outdated since SSB4 is only a few years old (less than 2 actually). The target system will be more powerful so they won't have to downgrade either. Unless there's a ton of PPC assembly to rewrite, I don't expect it should be too hard to do the basic port and build on that.

There will absolutely be new characters. Makes no sense not to add at least something. There's a few reasons for this.
For one, as Steve Youngblood pointed out, if this really is a launch game, it's because they want to incentivize people (including Wii U owners) to get the NX day 1. New characters in Smash Bros. guarantees the NX to at least have a good start. Wii U owners won't give a shit if it's just the same game. Also, Kimishima keeps stressing how well Smash DLC has sold. Do you really think they're going to abandon such a profitable venture? Characters in Smash are a huge seller.
Second, amiibo. The Smash Bros. line is far and away the most successful amiibo line, and I don't see that changing anytime soon. Adding new characters is such an easy way to get more amiibo sales, and it would be the easiest way to make sure NX has new amiibo on day 1.
Pichu is the least overrated Pokemon ever. I wish he wasn't a terrible clone in Melee.
Unless they add a character that I really want like Wonder Red, nothing will convince me to get Smash 4 again.
