I want a 2d Castlevania and Metroid.
Redesign, plain and simple. I will not own another one without a second analog stick if they choose to keep making games that need it.
It'd be nice if SE continued the FF remakes, VI especially.

There are no games out or being made that "need" it. In fact, very few games have been announced that even support dual analog control on 3DS.

This would be an ideal line up for me.

* Dragon Quest VII remake (I realize this is more of a Tokyo Game Show thing)
* New Zelda with new art style
* Pokemon Mystery Dungeon 3DS (Again, I'm aware Pokemon games are generally not E3 announcements)
* Anything Monolith Soft
* Confirmation that Chocobo Racing is still happening
All I mostly want is reassurance is that we'll get a variety of quality titles brought to the US, including games in niche genres. I'm a little worried that might not happen, so as long as that does, anything else they mention would be icing on the cake.

A proper Wario Land.
Wario Land Shake looked incredible (and was a great game), but it just didn't have the gameplay of the original three (and Virtual Boy). They need to expand on Wario Land 3, and really create an epic Wario game.
I mean hell, Wario Land 3 was essentially Wario crossed with Metroid.
I wonder if Conduit 3DS is still a thing. Come to think of it, HVS has been pretty quiet lately.Hopefully we'll see a surprising FPS or two.

I totally agree.
As much as I love both WL4 and Shake It!, they (especially the latter) veered away from the formula that made 2/3 such great (and original) experiences.
Re-inject the weirdness, return to a style that more closely resembles "Wario-metroidvania," and I'll be very happy.
