I'm hoping they don't spend much time on games like Pikmin 3 or Game and Wario. Nintendo needs to drop multiple bombs in the span of a very short time.
Here's what I'd like to see:
First 10 minutes: Start with a trailer for the new 3D Mario game and proceed to talk about it and demonstrate the unique gameplay. Have Miyamoto talk to the camera (not play) like they did for Luigi's Mansion 2 in a previous Direct. Hand it over to Iwata.
10-15 mins in: Talk about games already announced coming in the summer like Pikmin 3, Game and Wario, Wonderful 101, and whatever else I'm forgetting.
15-20 mins: Discuss the Summer System Update, as well as the new Wii U bundles. Discontinuation of the Basic Model and price drop to $299 for the Premium bundle, which is now known simply as Wii U - no "premium" branding. White "premium" console to be sold alongside the black one as well, and "deluxe digital promotion" not included anymore (to avoid consumer confusion as to what it is).
20-30 mins: 3DS focused segment. More gameplay of Zelda 3DS, Yoshi's Island 3DS, etc. and release dates. 1 new game announcement or so.
30-35 mins: Miiverse segment. 3DS is getting Miiverse TODAY, smart phone apps coming very soon. There's also room to discuss other apps like YouTube coming to 3DS.
35-40 mins: Sonic discussion. Talk about the partnership with Sega, show gameplay of Lost World and Olympic Games, and finally announce the third game in the partnership.
40-45 mins: Mario Kart. Show a trailer, give a release date, announce it's coming to Best Buy for you to try out.
45-55 mins: New game announcements. Retro's new game and everything else.
55-60 mins: Close it off with a Smash Bros. trailer and a surprise announcement that the demo will be available either at Best Buy or on the eShop, or both.
