When the OS is finally released, will there be video cards out on the market that have be tailored to use whatever it is they're replacing DirectX with? (If so, anyone know more about these cards and where I can read more about them?)

Pretty much, WGF(Windows Graphic Foundation) is the new DX that will launch with Longhorn, the biggest difference is that they linking DirectDraw and Direct3D together. Obviously it has a lot of the usual DX upgrades too.

Cards supporting it will probably appear in the next generation or the one after that, why would you want to find out about them?

I think you'll need a DX9 card to run Longhorn, at least that's what they're saying now. There may be some other hardware requirements though.
