No it was originally just an easy way for her to activate senketsu.

While it doesn't explain everything, she did bleed onto Senketsu in the first episode, remember? I think that extra blood allowed her to transform. Still, I'm not sure why Senketsu doesn't remember that.

Pretty sure the glove was just given to her by Aikuro as a quick way to draw blood. She could have used a lot of things. Later on, the glove seems to be retconned as just another part of the suit. I could have missed something tho.
