This seems like a logical idea for sorting out teams: there are 72 players currently in, divide by 4 (the number of players per team) equals 18. So that's 18 teams.
Take the 18 best players - all from Tier 1, and the remaining from Tier 2, and make them the captains/leaders of each team. That means the better players aren't going be together, so you're not going to get a situation like in school where people choose the best players because they know they'll win.
After that, I don't know. You could randomly draw teams, or you could work down the list, assigning players while moving through each team. So for example, you'd send Zoso Achilles to Team 1 and Mister Negative to Team 2 etc. while moving down.
