Depends on the weapon you use. If it's E scaling in Skill, dumping points into it won't gain you much additional damage, whereas if it has an B scaling in Skill, it's worth your points.So I've hit level 25 strength, should I drop points into skill now? Will that help?

I haven't started yet, but my copy should arrive today.
Is it practical to build toward using the Threaded Cane and Kirkhammer? These are the two coolest looking weapons I've seen, but it seems like they might be mutually exclusive, one scaling with skill and the other strength.
That's what I've been hearing. I'm probably going to pre-order dat thick Future Press Strat Guide instead.

I'm not so sure because the Threaded Cane uses Skill Stats while the Kirkhammer uses Strength stats. However, I'm sure you can build a hybrid STR/SKL build and interchange between both. There's nothing stopping you there except for the fact that you might need to level a lot more to use both effectively.
There are new lanterns in that area that can be lit and used, so if you get down those stairs and light the new lantern, yes, you can warp there.
One final question, for the end of the game;Is there a point of no return, or like Demon's and Dark Souls it's made very clear moving forward at a certain point is the final confrontation? I want to try and at least see and fight everything in my first playthrough, and I'm rather stumped at missing shortcuts. I've apparently skipped at least two boss fights, missed an NPC, and a key item for said boss fight. I hope the time of day doesn't play a role into that, because I may be screwed. ):
