I don't why I thought this was an online game only at first with no single-player campaign. I like the idea behind it.
I like the gameplay on PlayStation Youtube channel (and different to the press conference stage demo), you see them use the controller which is nice, and it's good quality:

Shooting looks great to me, as for movement, I would like some small lateral dogde moves, but otherwise it looks good. I'm wondering if you can build a character for speed, and what that plays like?

Looks like they drop generic ammo cubes instead.Really nice demo, learned a lot watching it. The size of the playable space, the dodge maneuvers. Looks good.
I just noticed that regular enemies don't appear to be dropping weapons, or at least they're not picking them up. Which makes me wonder about resource management; ammo, etc.

Thanks to a fella on the Bungie forums named nickgmon, he posted a video over 10 minutes long. It seem to be, at least from the description given, an alternative E3 trailer, with many new bits of footage and cool cinema like shots of planets we'll apparently get to visit.

How does mulitiplayer actually work?
From what has been said in the past, it'll work all in game. You leave the Tower in your ship and fly to an arena and duke it out in slayer/CTF types of game modes. This is based both on speculation and what I've heard from Bungie in interviews and the like.

From what I can gather, yes. (If you chose to play that way.)
I do not believe an online connection is required to play, but I may be wrong. Either way, they have said you can go solo and ditch the social aspects if you wish.
That sucks, I hope their servers hold up on launch day...

Given the type of game it is, thats fine with me.
Assuming it has good competitive MP I hope it has an offline custom game mode for tournaments, etc.

Have they confirmed a release window they're trying to target?

I'm guessing this will probably be a Fall 2014 title, but damn it would be so good to be playing this in March of next year.
There are reports that Activision doesn't want it colliding with CoD, so i'd expect early to mid 2014.

They're just rumors as far as I know. Activision hasn't came out and said that. But they only have a handful of franchises as you saw at this E3. They only showed Destiny, CoD and the new Skylanders game. That's it. So it's hard to believe that they'd want to place Destiny near CoD.
EDIT this is just speculation and guessing:I think it will still roughly "feel" like Halo with the gunplay (ADS aside of course). Probably player movement as well (even with the sliding added). For Run n Gun types from CoD it will feel slow.

Yeah, probably. But it won't play like classic Halos, i think. The closest will be Reach, if everyone had access to Sprint and two Armor Abilities, and no Halo-grenades (well, one class ability is probably grenade-like, we know the Warlock's Solar Flare works like a grenade (presumably)), with ADS.
I won't expect the game to be very competitive, not a game for those who look a large skill-gap.
Lame makes Destiny multiplayer not for me then. Had hoped they would give classes base skills and base gear kinda like how Guild wars pvp worked. So devs had an easier time to balance multiplayer.
At least that was what i was told, when a friend tried to convince me to drop wow for guildwars.
But never intended to get it for competitive multiplayer anyway.

Im in for the raids and dungeons just hope they have enough end game content or it will be like every mmo that hasn't fail.
I would be quite surprised if we get dungeons and raids in the traditional sense. Destiny doesn't really seems much like traditional mmo's ( is it even an actual mmo? ). Just look how different planetside 2, the only real fps mmo, is from traditional mmos like wow or rift.
